When it comes to cooking, there aren’t many kids who at one time or another don’t get curious about what goes on in the kitchen. It is not at all uncommon for a boy or girl to want to try their hand at preparing a meal or dessert, and parents should be very encouraging about it, as knowing how to cook is a life skill that the kids will use forever. It’s also a good time to teach them about food safety at home.

The great thing about teaching about food safety is that it allows you to work in so many other topics. For younger kids, spark their interest in chemistry by telling them about how surfactants, the things in cleaning products that allows them to penetrate dirt and grease and lift it away, work. Let them try cleaning the same grease with plain water, then with your favorite cleaning spray. If you have a microscope (you can get them fairly cheaply), swab a dirty countertop and let the kids see all the microscopic beasties crawling around in the goo.

For older kids, you can set up a biological experiment. Get some Petri dishes and nutrient agar, and then swab various kitchen surfaces before and after cleaning after a big cooking session. Help them identify the various colonies of bacteria that grow in the agar solution.

Pay attention to your kids’ interests and try to tailor the science activities to what they like. You are the best judge of what will work to get your message across.

Many kids won’t initially want to do the dirty work of cleaning up after they cook. The kitchen piled high with dirty pots and pans after a kid cooking session is one of the oldest jokes in the book, but it can be a thing of the past if you take the time to teach your children that cleaning up isn’t just the polite thing to do … practicing food safety at home could also save their lives and the lives of their diners!

Agar is an unbranched polysaccharide obtained from the cell walls of some species of red algae or seaweed. The word agar comes from the Malay word agar-agar (meaning jelly). It is also known as kanten or agal-agal (Ceylon agar). Chemically, agar is a polymer made up of subunits of the sugar galactose. Agar polysaccharides serve as the primary structural support for the algae’s cell walls. Dissolved in hot water and cooled, agar becomes gelatinous. Its chief use is as a culture medium for microbiological work. Other uses are as a laxative, a vegetarian gelatin substitute, a thickener for soups, in jellies, ice cream and Japanese desserts such as anmitsu, as a clarifying agent in brewing, and for paper sizing fabrics.

Nutrient agar is used throughout the world as a medium for the growth of bacteria and fungi, but not viruses (however, viruses are often grown in bacteria that are growing on agar). Though less than 1% of all existing bacteria can be grown successfully, the basic agar formula can be used to grow most of the microbes whose needs are known. More specific nutrient agars are available, because microbes can be picky. For example, blood agar, which is generally combined with horse blood, can be used to detect the presence of haemorrhagic micro-organisms such as E.coli O:157 H:7. Selective media is agar specially treated to apply a selective pressure to organisms growing on it — for example, to select for salt-tolerant, gram-positive, or gram-negative bacteria. To select for only gram negative organisms you would use MacConkey agar, which would also in turn tell you if the gram negative organism is a lactose fermenter or not indicated by red colonies instead of translucent (non- lactose fermenter).

There are several ways to make NA plates, all depending on what you have to start with. The well equipped person has clean and sterile empty petri plates, and a bottle of NA powder sitting on the shelf. That person needs only to follow the directions on the bottle

This is really not a project that requires a chicken little-esque response to. The sky isn’t falling, and the world will not end. Your mouth is full of bacteria. So is your entire body. The bacteria outnumber your human cells by 10 to 1 at least. And for whatever it is worth, in college (not 5th graders, I know) we cultured bacteria from much more terrible places in our body than our mouth and it was BSL-1.

Answer to the question follows, at the end I’ll recap biosafety levels.

You need a solidifying agent. Gelatine was used early on in microbiology to solidify agar. The problem with gelatine is that some bacteria degrade it, which is why agar is used now. But you should be able to try gelatine and see how it works.

The bacteria will need carbohydrates. You might want to try something like vegetable broth or maybe even V8? I know historically that these have been used as a component of growth media (one of the scientists in my lab makes some sort of V8 agar, he has cans of it on his bench).

Or you can try normal table sugar for this, since that’s something the bacteria in a human’s mouth encounter anyhow. Some bacteria cannot metabolize sucrose, however, just a detail to keep in mind.

The bacteria will also need some sort of protein in order to provide some amino acids and act as a nitrogen source. I’m not sure what to use for this, but something that dissolves and is high in protein.

To prepare it you’d want to mix everything together and then boil it until everything has dissolved. Let it cool a little then pour into whatever you’re using as petri dish surrogates, making sure that they have a cover of some sort. If possible, you might want to work out some way to “sterilize” the petri dish-thing, use some rubbing alcohol perhaps prior to adding your agar.

I’m not sure about eating it. In theory you’re just putting in stuff you would eat individually, so I don’t know that it would necessarily be bad to eat. But, a “slick” way to do the experiment would be to use some sort of sterilized q-tip or something to swab the persons teeth, then streak it across the plate, and do the same with the dog.

I’m pretty sure if working with sewage is BSL-1, then so is growing bacteria from your own mouth.

An agar plate is a sterile Petri dish that contains a growth medium (typically agar plus nutrients) used to culture microorganisms or small plants like the moss Physcomitrella patens.
Selective growth compounds may also be added to the media, such as antibiotics.
Individual microorganisms placed on the plate will grow into individual colonies, each a clone genetically identical to the individual ancestor organism (except for the low, unavoidable rate of mutation). Thus, the plate can be used either to estimate the concentration of organisms in a liquid culture or a suitable dilution of that culture using a colony counter, or to generate genetically pure cultures from a mixed culture of genetically different organisms, using a technique known as “streaking”. In this technique, a drop of the culture on the end of a thin, sterile loop of wire is streaked across the surface of the agar leaving organisms behind, a higher number at the beginning of the streak and a lower number at the end. At some point during a successful “streak”, the number of organisms deposited will be such that distinct individual colonies will grow in that area which may be removed for further culturing, using another sterile loop.

Like other growth media, the formulations of agar used in plates may be classified as either “defined” or “undefined”; defined medium is synthesized from individual chemicals required by the organism so that the exact molecular composition is known, whereas undefined medium is made from natural products such as yeast extract, where the precise composition is unknown.
Agar plates may be formulated as either permissive, with the intent of allowing the growth of whatever organisms are present, or restrictive or selective, with the intent of only allowing growth a particular subset of those organisms. This may take the form of a nutritional requirement, for instance providing a particular compound such as lactose as the only source of carbon and thereby selecting only organisms which can metabolize that compound, or by including a particular antibiotic or other substance in order to select only organisms which are resistant to that substance. This correlates to some degree with defined and undefined media; undefined media, made from natural products and containing an unknown combination of very many organic molecules, is typically more permissive in terms of supplying the needs of a wider variety of organisms, while defined media can be precisely tailored to select organisms with specific properties.
Agar plates may also be indicator plates, in which the organisms are not selected on the basis of growth, but are instead distinguished by a color change in some colonies, typically caused by the action of an enzyme on some compound added to the media.
Some commonly used agar plate types are:

Blood agar plate (BAP)
Contains mammalian blood (usually sheep or horse), typically at a concentration of 510%. BAP are an enriched, differential media used to isolate fastidious organisms and detect hemolytic activity. ?-hemolytic activity will show complete lysis of red blood cells surrounding colony. Examples include Streptococcus haemolyticus. ?-hemolysis will only partially lyse hemoglobin and will appear green. An example of this would be Streptococcus viridans. ?-hemolysis (or non-hemolytic) is the term referring to a lack of hemolytic activity.
Contains meat extract,tryptone, sodium chloride, and agar.
Chocolate agar (CHOC)
A type of blood agar plate in which the blood cells have been lysed by heating the cells to 56. Chocolate agar is used for growing fastidious (fussy) respiratory bacteria, such as Haemophilus influenzae. No chocolate is actually contained in the plate; it is named for the coloration only.
Thayer-Martin agar (TM)
Chocolate agar designed to isolate Neisseria gonorrhoeae.
